Kratos-SUPRA@UCI Update (11/23/2015)

The Kratos-SUPRA has been restored to operational in most common modes (XPS, depth profiling). During this maintenance period, the x-ray gun filament was replaced, the low count rate issues was fixed by cleaning out the restrictors on the cooling water lines. Currently, the pressure of SAC is at ~2E-09 torr. A long bake-out of the system is planned and the schedule will be updated. We recommend to keep your “degassing” samples in the loadlock chamber to pump down until the pressure reaches mid E-07 torr, before transferring to SAC

FEI XL-30 SEM computer (Sept 19-21) and EDS system upgrade

FEI Engineer will upgrade the computer and imaging software for FEI XL-30 SEM from Sept 19 to 21. The EDS system upgrade will follow up. I will conduct a quick training to get current XL-30 SEM users familar with the new software soon.  I am going to inform all current users to sign up for the training.

SEM short course:  How to Get Best Result from FEI/Philips XL-30 FESEM

 The SEM in MC2 is a field emission gun machine (FEI/Philips XL-30) with a resolution of 2nm. This SEM is equipped with E-T SE detector, solid state BSE detector and an EDS system, enabling users to obtain information about sample surface morphology and (qualitative and quantitative) chemical composition. To help users to take advantage of its full capabilities, a SEM short course “How to Get Best Result from FEI/Philips XL-30 FESEM” is scheduled on Sept 22, 2010. All current XL-30 SEM users and potential users are welcome to attend the one-day short course. The short course consists of 2.5hrs lecture (SEM principles and operations) in the morning and 2-4hrs (problem solving) demo in the afternoon. If you are interested in the short course, please email Dr. Zheng at jzheng@uci.edu in advance. If users have specific problems with their samples, they are welcome to inform Dr. Zheng in advance. A few users’ samples will be selected for the demo section.

Congratulations to Mercè Dalmau (Prof. Szu-Wen Wang’s group

Congratulations to Mercè Dalmau (Prof. Szu-Wen Wang’s group). She published a paper “pH-Triggered Disassembly in a Caged Protein Complex” in Biomacromolecules in Oct 2009 (vol 10, iss 12, p3199-3206), where MC2 CM-20 TEM was used to reveal the structure and disassembly of the caged protein complex (see figures above). The results are significant progress toward the use of the caged protein complex as a drug delivery vehicle. This paper and related TEM images have been featured recently in “Research Highlights” of Biopolymers (Vol. 93, no. 2, 2010).
This has been her third paper since Dec 2008. Other two papers are “Thermostability and Molecular Encapsulation within an Engineered Caged Protein Scaffold” published in Biotechnology and Bioengineering in Oct 2008 (vol 101, iss 4, p554-664) and “Design of a pH-Dependent Molecular Switch in a Caged Protein Platform” in Nano Letters in Jan 2009 (vol 9, iss 1, p160-166). The former was highlighted in B&B’s “Spotlight” Feature and TEM images were selected as cover photo in December 2008 (see picture below).
It is a great achievement to publish three high quality papers in such a short period of time!

XRD machine Problem 1/20/2010 fixed on 1/29/2010

Hi, XRD users,

 A X-ray diffractometer problem occurred this morning. After a brief check, I found that the problem might be associated with the X-ray generating system. I will talk with a service engineer to diagnose the reasons for the XRD problem  tomorrow. The XRD machine will be down till it is fixed. I will add comments to this news to keep you posted as the XRD status changes.

TEM CCD camera Jan 8/removed Feb 1/Loaner camera installed OK

TEM users:
TEM CCD camera lost some functions, but it is still working well at binning 1. Please use binning 1 instead of binning 2, 3 and 4. The image size for binning 1 is very big (11Mb for each image) , so please record TEM images only when necessary.

We will repair the camera and have it fully functional soon. If you have any questions, you may contact Dr. Zheng.
